Nicht-invasive Analysenmethoden der Schwingungsspektroskopie in der pharmazeutischen ForschungSiegfried Wartewig und Reinhard Neubert Institut für Angewandte Dermatopharmazie an der Martin-Luther-Universität Halle-Wittenberg, Halle/Saale Noninvasive Methods of Vibrational Spectroscopy in Pharmaceutical Research The basic principles of the Fourier transform (FT) technique and of the photoacoustic spectroscopy (PAS) are explained. To demonstrate the potential use of FT-Raman spectroscopy investigations on various topics are reported, namely the thermotropic properties of lipids, the differentiation of stratum corneum from healthy, undiseased atopic, and undiseased psoriatic skin, and the polymorphism of drugs. FTIR-PAS was applied to quantify the drug content in semisolid formulations and to monitor the drug penetration through membranes in real time. The step-scan modus of PAS offers the possibility for controllable depth profiling during the penetration process.
